If I saw this, my guess would be that the printer ran out of either
fleurons or asterisks. Maybe you could transcribe it as:

500 Signatures: [fleuron]8 A-D12 F4 ; (second leaf, [fleuron]2, signed
*2)

Hello all,
I am recording the signature statement for Joannis MiltonI Defensio
secunda pro populo Anglicano (Hagae-Comitum,1654) and the first 8 leaves
are signed: t.p. unsigned, *2, [fleuron]3-[fleuron]5, last 3 unsigned.
12mo format, the rest of the gatherings are singed A-D12 F4.
Not sure what to make of that first gathering.
Any suggestions?
